چکیده

Nine 5-bromodeoxyuridine-induced temperature-sensitive (ts) mutants were isolated from stocks of a syncytial plaque type mutant (syn) itself derived from the non-syncytial wild-type (syn +) of the GLASGOW strain i7 of herpes simplex virus type i. The nine ts mutants have been assigned to eight cistrons on the basis of different complementation tests. Non-syncytial revertants isolated from the original ts syn mutants were used in reciprocal three-factor crosses of the type: t sX syn x ts Y syn + and t sX syn + x ts Y syn. With recombination data from these crosses a linkage map has been constructed. The provisional map which locates nine cistrons is linear and spans about 25 recombination units. The regular occurrence of plaques with mixed syn/syn + morphology in the progeny from crosses is reported.

Qualitative complementation test for temperature-sensitive mutants of herpes simplex virus.

A simple, rapid, qualitative method for classifying temperature-sensitive mutants of herpes simplex virus into functional complementation groups has been developed. The positive reaction observed in this test reflects the ability of mutant pairs to interact by both complementation and recombination.

Method for induction of mutations in physically defined regions of the herpes simplex virus genome.

A procedure was developed for inducing mutations in isolated restriction enzyme fragments of herpes simplex virus type 1 (HSV-1) DNA with nitrous acid. The mutations were then transferred to the viral genome by genetic recombination during cotransfection of rabbit kidney cells with the mutagenized fragments and intact HSV-1 DNA. The HpaI restriction enzyme fragments LD, B, LG, I, and J were mut...

Replication Characteristics of Herpes Simplex Virus Type-1 (HSV-1) Recombinants in 3 Types of Tissue Cultures

A complication in the analysis of the role of ICP34.5 gene in the herpes simplex virus type-1 (HSV-1) lifecycle is the presence of overlapping antisense gene, open reading frame P (ORF P), which is also deleted in HSV-1 ICP34.5 negative mutants. A HSV-1 wild type strain (17+) ICP34.5/ORF P deletion mutant (1716) is totally avirulent in animal models and impaired in a number of in vitro function...

Genetic analysis of temperature-sensitive mutants which define the genes for the major herpes simplex virus type 2 DNA-binding protein and a new late function.

Eleven temperature-sensitive mutants of herpes simplex virus type 2 (HSV-2) exhibit overlapping patterns of complementation that define four functional groups. Recombination tests confirmed the assignment of mutants to complementation groups 1 through 4 and permitted the four groups to be ordered in an unambiguous linear array. Combined recombination and marker rescue tests (A. E. Spang, P. J. ...

Mutant analysis of herpes simplex virus-induced cell surface antigens: resistance to complement-mediated immune cytolysis.

BHK-21 cells infected with temperature-sensitive mutants of herpes simplex virus type 1 strain KOS representing 16 complementation groups were tested for susceptibility to complement-mediated immune cytolysis at permissive (34 degrees C) and nonpermissive (39 degrees C) temperatures. Only cells infected by mutants in complementation group E were resistant to immune cytolysis in a temperature-se...
